Shares of Verastem Oncology (VSTM) climbed 5% in after-hours trading Wednesday after the U.S. Food and Drug Administration granted Fast Track status to its experimental oral drug VS-7375, a move that could accelerate the development of a new treatment option for a subset of lung cancer patients with limited alternatives.

The Fast Track designation aims to speed the delivery of promising therapies for serious conditions by enabling more frequent interactions with regulators and potentially faster review timelines. For Verastem, the tag applies to patients with advanced non-small cell lung cancer (NSCLC) driven by the KRAS G12D mutation — a genetic alteration often described as a stuck “gas pedal” in cancer cells that drives uncontrolled growth.

Currently, no targeted therapy is approved in the U.S. specifically for KRAS G12D-mutated NSCLC, making the area a high-stakes race for biotech firms. The mutation occurs in roughly 5% of NSCLC cases — the most common form of lung cancer, accounting for 80% to 85% of all lung cancer diagnoses. That translates to more than 8,000 U.S. patients each year who are diagnosed with advanced or metastatic disease carrying this mutation, and they typically respond poorly to standard treatments and face a grim prognosis.

VS-7375 is an oral small molecule designed to block the mutated KRAS G12D protein in both its active and inactive states. Verastem believes this dual mechanism could offer a more durable tumor suppression compared with drugs that only target one conformational state. The company already holds Fast Track designation for the same drug in KRAS G12D-mutated pancreatic cancer.

The biotech is advancing VS-7375 through multiple clinical trials. An early-stage study is recruiting patients, with initial safety and efficacy data expected in the first half of 2026 and fuller results later that year. Verastem has also initiated a mid-stage, registration-directed trial focusing on advanced NSCLC patients who have received one or two prior lines of therapy. Notably, the trial includes patients with untreated brain metastases — a group often excluded from other studies.

On Stocktwits, retail sentiment around VSTM ticked up from ‘bearish’ to ‘bullish’ over the past 24 hours, while message volume jumped from ‘normal’ to ‘high.’ One user speculated that the company could become a buyout target, while another voiced confidence that VS-7375 could eventually become a drug on par with Merck’s blockbuster immunotherapy Keytruda, which revolutionized lung cancer treatment but works through a different mechanism.

Despite the after-hours pop, VSTM has fallen roughly 39% over the past 12 months, reflecting the broader challenges facing early-stage biotech stocks amid a volatile market and high cash-burn rates. The Fast Track designation provides a regulatory catalyst, but investors will remain focused on upcoming clinical data and the company’s ability to compete in the increasingly crowded KRAS inhibitor space.
